Trimethoprim

Trimethoprim is the dihydrofolate-reductase inhibitor in Bactrim / Septra and the prophylactic of choice for UTIs and Pneumocystis. The molecule is a 2,4-diaminopyrimidine tethered by a methylene bridge to 3,4,5-trimethoxybenzene. Two cuts open it: snap the methylene off the aryl side (Knoevenagel), then close the pyrimidine ring back onto guanidine.
